Protection of cholinesterase by ethanol against inhibition by organophosphates in vitro.

چکیده

Because many of the organophosphates used as anticholinesterases are insoluble in water, it is usual to dissolve them in water-miscible organic solvents in order to study their inhibition of the enzyme both in v&o and in vivo. Ethanol is commonly used for this purpose (1, 2), and, because it is an inhibitor of the enzyme, the control samples are treated with a similar level of the solvent. The extent of inhibition by the organophosphate is then obtained by reference to the alcohol-treated control, and the results are considered as corrected for the presence of alcohol and therefore comparable with results for water-soluble inhibitors obtained in ethanol-free solutions. In order to determine the validity of this assumption, experiments were carried out with several water-soluble organophosphates both with and without ethanol, and it was found that the solvent interferes strongly with the inhibition process.

Synthesis, X-Rays Analysis, Docking Study and Cholinesterase Inhibition Activity of 2,3-dihydroquinazolin-4(1H)-one Derivatives

In search of potent cholinesterase inhibitors, we have carried out the synthesis and biologically evaluation of various benzaldehyde based 2,3-dihydroquinazolin-4(1H)-one derivatives. In vitro assay results revealed that all the synthesized compounds showed activity against both enzymes (AChE and BChE) and in few cases, the inhibition activity was even higher than or comp...

بررسی اثر حفاظتی N ـ استیل سیستئین بر سطح گلوتاتیون کبد و آنزیم استیل کولین استراز خون در موش سوری، در مسمویت حاد ناشی ازدیازینون

Background and Purpose: The wide spread usage of pesticides for improving agricultural programs is a risk factor of acute and chronic human poisoning, especially farmers. Diazinon is one of the most common organophosphates insecticides that cause oxidative stress and lipid peroxidation. The mechanism of organophosphates is postulated to be non-irreversible anticholinesterase action. This study ...

Cholinesterases Enzymes Activities as Biomarkers of Farm Workers Exposed to Organophosphates in Two Communities of Khuzestan, Iran

Organophosphate (OPs) compounds are widely used in intensive agriculture to improve production, protect crops and control diseases vectors. The main mechanism of toxicity of OPs is the inhibition of the Cholinesterase enzymes. In this study the acetylcholinestrase (AChE), butyrylcholinesterase (BChE) and specific acetylcholinestrase (SAChE) activities as potential biomarkers of exposure to OPs ...
